Thermostable Chitinase Producing Bacterium from Ijen Hot Spring – Indonesia: Isolation, Identification, and Characterization

The high industrial demand for thermostable chitinase with new and desirable characteristics has led to the exploration of chitinolytic bacteria from extreme environments. Therefore, this study aimed isolate, screen, identify chitinase–producing Ijen hot spring, Indonesia. highest activity bacterium was identified by 16S rRNA gene sequencing its were confirmed morphological physiological analyses. Chitinase production selected under variation agitation aeration as well properties characterized afterward. Twelve bacterial colonies isolated screened their growth on Thermus colloidal chitin medium, in which used sole carbon source. Among these twelve isolates, isolate B2 showed activity. molecular, morphological, analyses that belonged Bacillus licheniformis . This produced a huge amount medium at 50 °C within 30 h. recorded 3 vvm rate 300 rpm speed. optimally active pH 7 temperature 55 evidently proofed due residual after several thermal stability tests.
